Touted as a high-octane emotional thriller, Karam is the story of one man's quest for retribution.

John (John Abraham) is an ace assassin who works for mob boss Captain (Bharat Dhabolkar). But one day in his life changes things forever. As he is in the midst of carrying out a cold blooded massacre of an entire family, he finds himself staring into the eyes of the little girl whose life is slowly ebbing away from her.

The realization of what he has become, hits John like a ton of bricks and he decides then and there to quit and start a new life with his wife Shalini (Priyanka Chopra). But the Karmic laws of life dictate something else for him.
Captain is facing a major threat from a rival don Yunus (Vishwajeet Pradhan) and after an attack on his life he decides to teach everyone a lesson by killing the city's top industrialist, top film producer, the cop backing Yunus, and lastly Yunus himself. But in order to carry out this plan he needs the help of his top hit man - John. He would be the only man who could make this plan work had he not already sworn never to pick up his gun again.

To coerce John into working for him again, Captain kidnaps John’s wife and holds her hostage, until he complies with the demands of killing the five targets within 36 hours.
In a fierce game of cat and mouse, John tries to fight the forces of nature and cut loose the strings of the puppet masters. He plots, double crosses and triple crosses everyone. He forms allegiances that will be broken. His friends turn into foes and the body count begins to grow. All the while Mumbai's toughest cop Wagh (Shiney Ahuja) is on his trail. Tormented by nightmares of a failure in his past, he is determined to bring John down when he realizes it was John who put a blot on his clean record years before.
